The Orica Bike-Exchange rider looked in control throughout the 163.2km ride from Monforte de Lemos to Luintra and launched a devastating attack with 3.6km remaining, en route to a convincing win over Luis-Leon Sanchez.
The Spaniard crossed the finish line 20 seconds adrift while Italy’s Fabio Felline had to settle for third place.
It was Yates’ second win of the season and comes just six weeks after he returned from a four-month suspension for testing positive for a banned substance, which was blamed on an “administrative error” made by his team doctor.
Chris Froome finished safely to stay third overall, 32 seconds behind race leader Darwin Atapuma of BMC. Spain’s Alejandro Valverde is currently sitting second going into Stage 7, a 158.5km trek from Maceda to Puebla de Sanabria.
